Abstract

Content of enhancers and links to transcription factors ; orthography, syntax and grammar. Genomic topology of enhancers.

In this fourth lesson, the ways in which enhancers can bind transcription factors are described, along with the notion of consensus DNA sequences. The modes of distribution of such sequences within a single enhancer are then described. Next, the use of a convolutional neural network is presented, to find enhancer sequences simply by reading DNA, after deep learning. These promising results pave the way for the production of syntheticenhancer sequences with desired properties for a specific purpose.

In the second part of the lesson, an examination of different genomicenhancertopologies is proposed, i.e. how multipleenhancers are arranged in regulatory landscapes. Some recurrent topologies are presented, with examples of critical genomic loci in mammalian development.
