Abstract

Detection ofenhancers by epigenetic and chromatin structure approaches, multiome approach, mechanisms of enhancer function to initiate transcription of a target gene.

In this third lesson, the different approaches to epigenetic profiling are explained, as well as their relationships with the different functional states of chromatin. Current methods involving the capture of chromosome interactions with the aim of establishing chromatin topology are then described.

Next, the potential mechanisms used to bring enhancerscloser to their target promoters are discussed ( loop extrusion model ) and integrated into an overall (simplified) presentation of the events leading to the initiation of transcription of a target gene by successive aggregation of factors. The cellular specificity of this process lies in the factors associated with theenhancer, i.e. its DNA sequence.
