Types of regulatory landscapes. Analysis of chromatin architectures at the single-cell level using super-resolution microscopy.
In this fifth lesson, different types of topological organization ofenhancers are described, in particular those allowing pleiotropic expression of the target gene. How can different enhancers arranged in the same chromatin domain be mobilized in different combinations, with distinct spatio-temporal specificities ?
Next, the question of single-cell analysis of chromatin architectures is raised, since current analyses deal with a set of cells and therefore give a combined representation of a set of mutually exclusive possibilities for a given allele. A study is presented that uses super-resolution microscopy (STORM) to establish the number of possible configurations and their representativities at the Sonic hedgehog (Shh) gene locus .
